Abstract

Background: Tumor necrosis factor alpha (TNFα) is a 17 kDa, an important soluble pro-inflammatory cytokine, which is involved in some tissue dysfunctions, including thyroid and liver tissue. In spite of its role in thyroid and tissue damage separately, the relationship between this factor and these two disorders has not been clarified. The aim of the present study was to evaluate liver biochemical parameters and TNFα in hypothyroid patients compared to euthyroid subjects.
Methods: To achieve this purpose, samples were transferred into tubes without anticoagulants and then centrifuged immediately to separate the serum. All markers in the serum were measured using commercial kits, including T3, T4, TSH, and TNFα, which were detected using the ELISA method. Liver function tests, including albumin, total bilirubin, and total protein were measured by spectroscopy and the colorimetric method, respectively. In addition, AST, ALT, ALP, and GGT were detected using enzymatic methods.
Results: Our results showed that the level of TNFα in hypothyroid patients was significantly higher than that in normal individuals (P = 0.009). TNFα had a significantly positive correlation with TSH and T3 but a negative correlation with T4. Furthermore, AST, ALT, and GGT had a positive correlation with TSH and a negative correlation with albumin, total protein, and total bilirubin. These correlations were insignificant (P < 0.05).
Conclusion: According to our data, the positive correlation of TSH with both TNFα and liver function tests may indicate a relationship between thyroid and liver function with each other.

Abstract

Background: Diabetic nephropathy is a major cause of kidney failure. The current study aimed to measure serum and urine albumin, as well as other biochemical markers, in individuals with type 2 diabetic nephropathy and to identify rapid diagnostic approaches in these patients.
Methods: This case-control study included 40 patients with diabetic nephropathy and 40 diabetic patients without nephropathy as the control group. Clinical parameters were assessed. After biochemical analysis and based on the ratio of random urine albumin to creatinine, patients were divided into two groups: those without albuminuria and those with microalbuminuria. SPSS version 18 software was used to analyze the results.
Results: In diabetic patients, the levels of urea, creatinine, and uric acid were significantly higher compared with the control group and were 64.68±46.84, 2.01±1.82, and 5.35±2.23, respectively. The average serum albumin content in diabetic patients with nephropathy was notably lower than that in patients without nephropathy (3.74±0.88 and 4.35±0.28, respectively) (p < 0.05). In patients with diabetic nephropathy, urinary albumin had a direct relationship with serum creatinine level (r=0.347, p=0.028). In the control group, serum albumin status could be assessed by measuring creatinine (r = -0.305, p = 0.056), urea (r = -0.333, p = 0.036), and HbA1C (r = -0.376, p = 0.017).
Conclusion: The results of this study revealed that high urinary albumin levels in patients with diabetic nephropathy have a direct association with serum creatinine levels, indicating a useful marker for evaluating nephropathy patients with albuminuria. However, further studies with larger numbers of participants are required.

Abstract

Abstract
Background: Biofilm formation in Staphylococcus aureus (S. aureus), mediated by the ica operon, is a key virulence factor. This study examined how different glucose-supplemented broth culture media influence biofilm production and ica gene expression in S. aureus.
Methods: The phenotypic ability to adhere to a polystyrene surface and to produce slime layer were evaluated using microtiter plate test (MtP) and Congo red tube test, respectively. Using PCR, the presence of intercellular adhesion (ica) locus in S. aureus strains was confirmed and subsequently, quantitative real-time RT-PCR was performed to investigate transcription of icaA in various media including Tryptic soy broth (TSB), Brain-heart infusion broth (BHIB), (Nutrient broth) NB and (Muller-Hinton broth) MHB contained 0, 0.25, 0.5, 1 and 2% glucose.
Results: Our results showed that although all of the studied strains adhered to the wells of polystyrene microtiter plates, the optimum rate of biofilm formation was observed for TSB medium contained 1% glucose, but biofilm formation was not significantly different in NB, MHB and BHIB media. Supplementation of all media with 1% glucose led to the highest production of biofilm formation and in all media transcription of icaA was increased with glucose addition to one present.
Conclusions: The results of the present study indicated that TSB medium supplemented with 1% glucose was the most appropriate medium for evaluation of biofilm formation by S. aureus isolates.
